While not advocating the weight weenie school of cycling size, in this case, weight definitely matters as the physics of rotating mass tells you that the lighter you can get it the faster you can turn it.

All you need to consider whether it is worthwhile having a dynohub to give options for lighting.
As far as where to start, look at ready built wheels and see how they are constructed and where manufactures put their money or save money in the process.
Finally, consider issues associated with use and the ease of repairing/maintenance in an emergency. Having the finest wheel in the world, for your purposes, is great but what if you have chosen components that are not readily available and will stop your ride due to a failure. Being stuck in the boondocks or even having a long walk to get assistance is not fun!!!
