Even with index shifting I can't see any problem tbh. Shimano shifters are able to push past the last indexed position so with the sprocket only 0.5mm further outwards than where a 6 speed would be I can't see that as an issue.
Regarding fitting the last sprocket on, given the very minute variance there will still be enough threading on the freehub to be able to tighten it down okay/safely.
If you pose the Q on RB you'd get a definitive answer.
Worse case scenario you get a 6 speed top sprocket or dremell 0.5mm off the spacer
