Have twice had similar symptoms and kept on fiddling, only for the cable to fray and snap inside the STI. On old shimano shifters it's a ball ache to get out, newer ones now have a removable part of the lever housing to help.
It happened again to me this week, with cables that were installed with a brand new group set less than 9 months ago.
